As a peptide, semaglutide is sensitive to conditions that break amide bonds or modify side chains. Deamidation of asparagine and glutamine residues, oxidation of methionine and tryptophan, and non-covalent aggregation are the main degradation routes described in published stability work. Rate depends strongly on pH, buffer species, ionic strength, temperature and exposure to light. Formulators therefore choose a defined solution pH and often add excipients such as phosphate buffer, propylene glycol and phenol, each of which plays a separate role in pH control, tonicity or preservation.
Storage guidance for the finished injectable product distinguishes the unused state from the in-use state. Before first use, pens are kept refrigerated between 2 and 8 degrees Celsius, protected from light, and never frozen, since freezing can disrupt the peptide or the device. After first use, label instructions in several markets permit storage at room temperature up to about 30 degrees Celsius for a limited number of days. Solid research-grade material is normally held at or below minus 20 degrees Celsius, often with desiccant, and allowed to equilibrate before opening.
The primary target is the GLP-1 receptor, a class B G protein-coupled receptor expressed on pancreatic beta cells, in the gut, and in several brain regions. Receptor activation raises intracellular cyclic AMP, which potentiates glucose-dependent insulin secretion and lowers glucagon release when blood glucose is elevated. Signalling in the hypothalamus and brainstem is associated with reduced appetite and slower gastric emptying. Because the insulinotropic effect depends on prevailing glucose levels, the hypoglycaemic risk of the peptide alone is described as low in most study settings. The relative contribution of peripheral and central actions remains an active research question.
Large randomised trials in adults with type 2 diabetes and in adults with obesity have reported reductions in body weight and improvements in several cardiovascular risk markers. One outcome trial found a lower incidence of major adverse cardiovascular events in participants with diabetes and established cardiovascular disease. Gastrointestinal effects such as nausea and vomiting are the most frequently reported adverse events and often diminish over time. Changes in lean body mass during weight loss are an area of ongoing investigation. Effects in adolescents and in pregnancy are less well characterised, and current labelling advises against use during pregnancy.
Semaglutide is a synthetic peptide analogue of glucagon-like peptide-1, a gut hormone released after nutrient intake. The molecule contains 31 amino acid residues and differs from the native sequence at several positions. A non-natural residue at position eight resists the enzyme that normally truncates the hormone, while a lysine-linked fatty diacid side chain promotes binding to serum albumin. These two modifications extend the circulating half-life from minutes to roughly one week. The peptide is produced by solid-phase synthesis followed by selective acylation, and its identity and purity are confirmed by spectrometric and chromatographic techniques.

Handling practices center on minimizing contamination and adsorption. Lyophilized peptide tends to accumulate static charge, so weighing is done with antistatic measures and calibrated balances. Reconstitution with appropriate solvent should be gentle, avoiding vigorous vortexing that generates foam and shear. Solutions are typically aliquoted before freezing to reduce repeated temperature cycling. Personal protective equipment and a fume hood are standard for powder handling.
Reconstituted solutions are less stable than the dry powder, and stability depends on concentration, pH, buffer composition, and container material. Low-protein-binding tubes reduce loss of peptide to plastic surfaces. Some researchers add a carrier protein to limit adsorption at low concentrations. The exact shelf life of a given solution is best determined empirically through a stability study rather than assumed from general guidance, because published data cover only a limited set of conditions.
Storage conditions for semaglutide depend heavily on the presentation. Lyophilized research powder is generally kept at two to eight degrees Celsius in a sealed container, protected from light and moisture. Manufacturer labeling for finished injectable products specifies refrigeration before first use, with defined in-use periods at room temperature afterward. The oral tablet form is stored at controlled room temperature and is more tolerant of short excursions. Temperature excursions should be documented rather than inferred.
Stability studies examine how temperature, pH, and moisture influence degradation rates over time. In aqueous solution, hydrolysis and deamidation accelerate as pH moves away from mildly acidic conditions. Light exposure and residual metal ions can also trigger oxidation of susceptible residues. Accelerated aging at elevated temperature is used to estimate shelf life, though extrapolation to room temperature carries uncertainty because individual degradation pathways do not always scale predictably.
Lyophilized semaglutide powder is typically held at minus twenty degrees Celsius for long-term storage. At that temperature, solid-state degradation reactions proceed slowly and the peptide remains intact for extended periods. Repeated freeze-thaw cycles are best avoided because they promote aggregation and can shift the proportion of monomeric peptide present. Working aliquots are often prepared so that each portion is thawed only once, and desiccant is placed inside the vial to limit moisture uptake.
GLP-1 receptors are expressed on pancreatic beta cells, in the gut, and in several brain regions. Receptor activation raises cyclic AMP, enhances glucose-dependent insulin secretion, and suppresses glucagon release when blood glucose is high. Effects on gastric emptying and on hypothalamic appetite circuits reduce energy intake. Because insulin release remains glucose-dependent, the risk of hypoglycemia is low when the drug is used alone. The precise contribution of each pathway to body weight change in humans remains an area of active investigation.
Clinical studies of semaglutide generally measure glycated hemoglobin, fasting plasma glucose, body weight, and composite cardiovascular endpoints. The SUSTAIN program enrolled adults with type 2 diabetes, while the STEP program focused on obesity without diabetes. Administration follows a stepwise escalation schedule designed to limit gastrointestinal effects during the first weeks. Reported outcomes include mean percentage weight change, the proportion of participants reaching defined weight-loss thresholds, and rates of nausea, vomiting, and diarrhea. Long-term data on durability after treatment stops are still limited and remain a topic of ongoing research.
Semaglutide is a synthetic peptide analog of human glucagon-like peptide-1, developed by Novo Nordisk and first approved in 2017 for type 2 diabetes. It belongs to the incretin mimetic class, a group of agents that reproduce the glucose-dependent actions of endogenous GLP-1. The molecule was engineered to resist degradation by dipeptidyl peptidase-4 and to bind serum albumin, extending its half-life from minutes to roughly one week. Approval for chronic weight management followed in 2021, based on large cardiovascular and obesity outcome trials.
Clinical development of this compound followed earlier short-acting GLP-1 analogues that required frequent injection. Once-weekly subcutaneous formulations entered use after 2017, and an oral formulation using a permeation enhancer later became available. The oral version pairs the peptide with sodium N-(8-[2-hydroxybenzoyl] amino) caprylate, a carrier that improves uptake across the gastric epithelium. Interest has expanded from glycaemic control into weight management and metabolic liver disease. Regulatory status and approved indications differ between countries, and the product remains subject to ongoing safety monitoring.
Semaglutide is a synthetic peptide that acts as an agonist at the glucagon-like peptide-1 receptor. It is a structural analogue of human GLP-1(7-37), modified to resist enzymatic degradation by dipeptidyl peptidase-4. The peptide backbone contains alpha-aminoisobutyric acid at position 8, a substitution that stabilises the helix and slows cleavage. A fatty diacid side chain attached through a linker at lysine 34 promotes binding to serum albumin, which extends the circulating half-life. These two modifications together allow less frequent administration than native GLP-1 requires.
== Other uses == The non-covalent bond formed between biotin and avidin or streptavidin has a binding affinity that is higher than most antigen and antibody bonds and approaches the strength of a covalent bond. This very tight binding makes labeling proteins with biotin a useful tool for applications such as affinity chromatography using immobilized avidin or streptavidin to separate the biotinylated protein from a mixture of other proteins and biochemicals. Biotinylated protein such as biotinylated bovine serum albumin (BSA) is used in solid-phase assays as a coating on the well surface in multiwell assay plates. Biotinylation of red blood cells has been used as a means of determining total blood volume without the use of radiolabels such as chromium 51, allowing volume determinations in low birth weight infants and pregnant women who could not otherwise be exposed to the required doses of radioactivity. Furthermore, biotinylation of MHC molecules to create MHC multimers has become a useful tool for identifying and isolating antigen-specific T-cell populations. More recently, in vivo protein biotinylation was developed to study protein-protein interactions and proximity in living cells
Increasing the Mg2+ concentration leads to an increase in the equilibrium constants for the aminoacyl-tRNA synthetases' reactions. Although this trend was seen in both class I and class II synthetases, the magnesium dependence for the two classes are very distinct. Class II synthetases have two or (more frequently) three Mg2+ ions, while class I only requires one Mg2+ ion. Beside their lack of overall sequence and structure similarity, class I and class II synthetases feature different ATP recognition mechanisms. While class I binds via interactions mediated by backbone hydrogen bonds, class II uses a pair of arginine residues to establish salt bridges to its ATP ligand. This oppositional implementation is manifested in two structural motifs, the Backbone Brackets and Arginine Tweezers, which are observable in all class I and class II structures, respectively. The high structural conservation of these motifs suggest that they must have been present since ancient times.

Marley's works feature animals, insects, fossils, minerals, botanicals, bones, and sea life. The animals, minerals, and other animal artifacts featured in his works are typically set in frames with white backgrounds. Marley's work contrasts traditional taxidermy by preserving the entire organism, not just the skin. Animals and insects he has featured in his works include: pythons, venomous reptiles, green mambas, Gaboon vipers, Canebreak rattle snakes, Chrysochus beetles, stag beetles, Delias bufferflies, Eupholus weevils, baby alligators, mustached parakeets, great blue turacos, military macaws, and Boa constrictors. Marley's artistic philosophy is motivated by aesthetics, telling scientific stories of biological diversity, and focusing on either one or two features in the mosaic for the general public to better receive. While abiding by local municipalities and their regulations, Marley has developed a network of zoos, aquariums, catchers, lepidopterists, coleopterists, entomologists, and breeders to legally supply him with animal artifacts to be used in his work. Notably, he doesn't buy specimens from hunters, and uses reclaimed specimens that have been caught as fishing bycatch or died by natural causes. He has also developed a novel freeze-drying method to preserve the animals, which contrasts the typical method of preserving animals in liquid. The insects shown in Marley's works are harvested by indigenous communities and sold to help support the local economies.

Determined to flee Wellington Wells with Gwen, Sally plans to steal General Byng's personal motorboat hidden near his military base. She convinces Dr. Helen Faraday (Samantha Lee) to create a new engine for the boat but fails to steal the key, getting knocked out by a sleeping dart trap. She awakens to find herself with General Byng in his safehouse; Byng intends to keep Sally imprisoned there until the problems in Wellington Wells are resolved while sending Gwen away to the mainland. Sally refuses, fights Byng, takes the key to the boat, and locks him inside the safehouse. Sally sneaks Gwen to the boat at night and rides it out of Wellington Wells. Ollie's Story: Ollie Starkey (Allan James Cooke) is a former British Army soldier, who lives as a recluse at his fortified hideout in the Garden District. His only company is a talking hallucination of his daughter, Margaret (Eloise Webb), killed years ago during the Very Bad Thing. After helping Arthur along his journey and having his hideout destroyed by Wastrels, he finds his former commander, General Byng. He informs him of the papier-mâché tanks Arthur had discovered. Byng reveals that he knew about the fake tanks but remained quiet to avoid a rebellion, which he doubts would have been successful. He also reveals that Ollie knew about the tanks as well, having served as Byng's orderly at the time. Unable to remember, Ollie leaves to confront Byng's daughter, Victoria.
Nayib Armando Bukele Ortez (born 24 July 1981) is a Salvadoran politician who has served as the 43rd president of El Salvador since 2019. In 1999, Bukele established an advertising company and worked at another owned by his father, Armando Bukele Kattán. Both companies advertised election campaigns for the Farabundo Martí National Liberation Front (FMLN) political party. Bukele entered politics in 2011. In 2012, he joined the FMLN and was elected mayor of Nuevo Cuscatlán. Bukele served until his 2015 election as Mayor of San Salvador, where he served until 2018. In 2017, Bukele was ousted from the FMLN. He founded the Nuevas Ideas political party shortly afterward and pursued a presidential campaign in 2019. After the Supreme Electoral Court (TSE) refused to register his party, Bukele ran for president with the Grand Alliance for National Unity (GANA) and won with 53 percent of the vote. In July 2019, Bukele implemented the Territorial Control Plan to reduce El Salvador's 2019 homicide rate of 38 per 100,000 people. Homicides fell by 50 percent during Bukele's first year in office. After 87 people were killed by gangs over one weekend in March 2022, Bukele initiated a nationwide crackdown on gangs, resulting in the arrests of over 85,000 people with alleged gang affiliations by December 2024. El Salvador's homicide rate officially decreased to 1.9 homicides per 100,000 in 2024, one of the lowest in the Americas, although the factual accuracy has been contested.
Draft scripts for Half-Life 2 indicate that this would have been shown in an introductory segment to the game carried out through a series of projector slides. One of the slides would have shown Breen at the foot of a tower wearing a headset linked directly to it, with arms held wide and speaking to the skies. Breen is alerted to the return of Gordon Freeman in Half-Life 2 when Gordon is temporarily teleported, by accident, to his office in the Citadel. Dr. Breen informs the Combine and immediately dispatches the forces at his disposal to capture Freeman and break the associated Resistance movement in City 17. During Gordon Freeman's raid on the Citadel, Freeman is temporarily in the custody of Breen, until Judith Mossman turns against the administrator. During this period, Breen makes a very notable statement while in the presence of Alyx Vance and her father, Eli (who are also in his custody). He claims that Gordon "has proven a fine pawn to those who control him." He also comments that Gordon's services are "open to the highest bidder," and says he would understand if Gordon doesn't want to discuss it in front of his friends. These remarks imply that Breen may be aware of the mysterious G-Man and his influence over Freeman. It was also mentioned in one of the "Breencasts" to the Sector Seventeen Overwatch in Nova Prospekt; "I have good reason to believe that in the intervening years, he was in a state that precluded further development of covert skills." When Judith Mossman frees Gordon Freeman and Alyx Vance in his office, Dr.
Plant hormones (or phytohormones) are signal molecules, produced within plants, that occur in extremely low concentrations. Plant hormones control all aspects of plant growth and development, including embryogenesis, the regulation of organ size, pathogen defense, stress tolerance and reproductive development. Unlike in animals (in which hormone production is restricted to specialized glands) each plant cell is capable of producing hormones. Went and Thimann coined the term "phytohormone" and used it in the title of their 1937 book. Phytohormones occur across the plant kingdom, and even in algae, where they have similar functions to those seen in vascular plants ("higher plants"). Some phytohormones also occur in microorganisms, such as unicellular fungi and bacteria, however in these cases they do not play a hormonal role and can better be regarded as secondary metabolites.
k is the Boltzmann constant, T is absolute temperature and F is the force exerted on a single particle by the force field. This shows how the characteristic elevation value is inversely dependent to the force applied. Therefore, F governs the separation process. Hence, by varying the field strength the separation can be controlled to achieve optimal levels. The velocity V of a cloud of molecules is simply the average velocity of an exponential distribution embedded in a parabolic flow profile. Retention time, tr can be written as:

=== Colors of lanthanide complexes === Superficially lanthanide complexes are similar to those of the transition metals in that some are colored. However, for the common Ln3+ ions (Ln = lanthanide) the colors are all pale, and hardly influenced by the nature of the ligand. The colors are due to 4f electron transitions. As the 4f orbitals in lanthanides are "buried" in the xenon core and shielded from the ligand by the 5s and 5p orbitals they are therefore not influenced by the ligands to any great extent leading to a much smaller crystal field splitting than in the transition metals. The absorption spectra of an Ln3+ ion approximates to that of the free ion where the electronic states are described by spin-orbit coupling. This contrasts to the transition metals where the ground state is split by the crystal field. Absorptions for Ln3+ are weak as electric dipole transitions are parity forbidden (Laporte forbidden) but can gain intensity due to the effect of a low-symmetry ligand field or mixing with higher electronic states (e.g. d orbitals). f-f absorption bands are extremely sharp which contrasts with those observed for transition metals which generally have broad bands. This can lead to extremely unusual effects, such as significant color changes under different forms of lighting.
=== Mechanism of action === Aceclidine is a selective muscarinic acetylcholine receptor agonist used primarily in ophthalmology. Its mechanism of action involves binding preferentially to muscarinic receptors on the iris sphincter muscle, leading to pupillary constriction (miosis) with minimal stimulation of the ciliary muscle. Unlike non-selective miotic agents such as pilocarpine, aceclidine's selectivity results in a pronounced reduction in pupil size without significant induction of accommodation or myopic shift, thereby minimizing side effects like lens thickening or accommodative spasm. This targeted action is leveraged in the treatment of presbyopia, where the drug induces a “pinhole” effect: by creating a small, stable pupil, aceclidine increases the depth of focus and improves near vision while maintaining distance visual acuity. Its limited activity on the ciliary muscle distinguishes it from other cholinergic agonists and offers a favorable safety profile, with a reduced risk of retina or vitreous traction and fewer accommodative disturbances.
=== Other ventures === Around 2013, Newell co-founded and funded Foundry10, an educational research organization. In 2018, Newell donated $20 million to the American artificial intelligence company OpenAI and acted as an informal advisor. In 2022, with Philip Sabes, Newell co-founded the neuroscience company Starfish Neuroscience to develop neural interfaces. In May 2025, Starfish announced that their first chip would be released late that year. Newell is the owner of the marine research organization Inkfish, which owns several ships and submarines. In November 2022, Inkfish purchased the Hadal Exploration System, a private deep-sea exploration platform, from the undersea explorer Victor Vescovo. In August 2025, Newell acquired the privately owned yacht manufacturer Oceanco.
=== EU risk assessments === In 2002 the EU Scientific Committee on Veterinary Measures relating to Public Health (SCVPH) claimed that the use of beef growth hormones posed a potential health risk, and in 2003 the EU enacted Directive 2003/74/EC to amend its ban, but the US and Canada objected that the EU had not met WTO standards for scientific risk assessment. The EC claimed that the hormones remain in the tissue, specifically 17-beta estradiol. However, the EC found no clear link to health risks in humans for the other five hormones. The EC reported that high amounts of hormones had been found in areas around dense cattle lots. This increase in hormones in groundwater affected waterways and nearby wild fish. Hormone contamination of North American waterways would not, however, have any health impacts on European consumers.
=== 1947–1969: Unions and foundations === The Nederlandse Radio Unie (Netherlands Radio Union; NRU) was established in 1947. After several failed attempts to create a public broadcasting system and link up with a national station, the NRU was created as a union of broadcasting associations that provided operational support. The associations were responsible for their output, but studios, orchestras, and outside broadcast facilities were managed by the NRU. Weekly radio plays were also the domain of the NRU and would run until 1986. The NRU became the Dutch founding member of the European Broadcasting Union in 1950. Meanwhile, the Nederlandse Televisie Stichting (Netherlands Television Foundation; NTS) was created in 1951, two years after public television returned to the airwaves. The NTS served as a similar organisation to the NRU, in that broadcast and transmission facilities were supplied to member associations for making programmes. It was not until 1956 that the NTS itself produced its first programme, a news bulletin called the NTS Journaal. This was followed by a sports round-up, Sport in Beeld (Sports Illustrated) in 1959, and 1967 Langs de Lijn (Along the Line), a joint production of several broadcasting associations. On 15 October 1967, Willem Leonard Oltmans interviewed the de Mohrenschildts for the NTS, which resulted in a 40-minute film that was the only full-length filmed interview of George de Mohrenschildt. However, the film, which was kept at Hilversum, disappeared in 1975.

Ice formation concentrates solutes and can mechanically stress the peptide or damage the delivery device. Thawing afterwards may leave aggregates that are not visible to the eye. Refrigeration keeps the solution above its freezing point while slowing chemical degradation.
Size-exclusion chromatography is the most common technique, separating monomer from dimer and higher-order species by hydrodynamic volume. It is usually paired with ultraviolet detection and, for trace work, with multi-angle light scattering. Results depend on mobile-phase conditions, so method details matter when comparing datasets.
The detailed degradation profile under real-world temperature excursions and repeated handling is not fully described in the open literature. Relative contributions of photo-degradation versus thermal pathways are also incompletely mapped. Whether specific impurity species carry different biological activity is an open question rather than an established finding.
Native GLP-1 is degraded within minutes by dipeptidyl peptidase-4 and neutral endopeptidases. Semaglutide carries a non-natural amino acid at position eight that blocks that cleavage, and a fatty diacid side chain that binds albumin. The result is a much longer duration of action than the native hormone.
