Reverse-phase high-performance liquid chromatography with ultraviolet detection near 214 or 280 nanometres is widely used to assess purity and to resolve related impurities. Liquid chromatography coupled to mass spectrometry confirms identity through the protonated molecular ion and fragment ions formed in tandem experiments. Capillary electrophoresis and peptide mapping after enzymatic digestion supply complementary information on charge variants and modification sites. Circular dichroism and nuclear magnetic resonance can report on secondary structure in solution. Absolute quantification usually depends on an external standard, and reported purity depends on the detection wavelength and integration parameters chosen.
Lyophilised material appears as a white to off-white cake or powder that is hygroscopic, and containers are usually equilibrated to room temperature before opening to limit condensation. Dissolution is performed in water, phosphate-buffered saline, or a mildly alkaline buffer, since solubility rises above neutral pH. Gentle inversion or low-speed mixing is preferred, because vigorous vortexing can promote surface denaturation and aggregation. Complete dissolution may require several minutes, and brief sonication is sometimes applied. Passing the solution through a 0.22 micrometre membrane removes particulates but does not by itself sterilise the liquid.
Storage at minus 20 degrees Celsius or lower in a desiccated container preserves the peptide for extended periods, while working solutions are commonly held at two to eight degrees Celsius for short intervals. Light exposure and repeated freeze-thaw cycles accelerate degradation, so dividing material into single-use aliquots is generally recommended. Adsorption to glass and plastic surfaces can lower the measured concentration of dilute solutions, particularly below one milligram per millilitre. The degradation routes most often reported for GLP-1 analogues are deamidation, methionine oxidation, and backbone hydrolysis. Relative rates under specific conditions are frequently described only for individual formulations.
Reported molecular weight is approximately 4113.6 daltons for the free base, and the peptide is supplied as a lyophilized powder or in buffered liquid form depending on the intended use. It is freely soluble in water when formulated with appropriate excipients, though the unconjugated peptide shows limited stability at neutral pH over long periods. Analytical characterization typically relies on reversed-phase high-performance liquid chromatography and mass spectrometry. Purity specifications for research-grade material commonly exceed ninety-five percent by area. Isotopic and impurity profiles differ between suppliers.
Two principal therapeutic variants exist under separate regulatory filings, one indicated for glycemic control in type 2 diabetes and one for chronic weight management. Both use the same active molecule; differences lie in formulation strength, titration schedule, and labeling. Regulatory agencies in the United States and European Union approved injectable forms in 2017 and 2018 respectively. An oral tablet formulation received approval later, using a carrier molecule to enhance absorption across the gastric epithelium. Labeling differs by jurisdiction and by indication.

Semaglutide is a synthetic peptide analogue of glucagon-like peptide-1, a gut hormone released after nutrient intake. The molecule contains 31 amino acid residues and differs from the native sequence at several positions. A non-natural residue at position eight resists the enzyme that normally truncates the hormone, while a lysine-linked fatty diacid side chain promotes binding to serum albumin. These two modifications extend the circulating half-life from minutes to roughly one week. The peptide is produced by solid-phase synthesis followed by selective acylation, and its identity and purity are confirmed by spectrometric and chromatographic techniques.
The primary target is the GLP-1 receptor, a class B G protein-coupled receptor expressed on pancreatic beta cells, in the gut, and in several brain regions. Receptor activation raises intracellular cyclic AMP, which potentiates glucose-dependent insulin secretion and lowers glucagon release when blood glucose is elevated. Signalling in the hypothalamus and brainstem is associated with reduced appetite and slower gastric emptying. Because the insulinotropic effect depends on prevailing glucose levels, the hypoglycaemic risk of the peptide alone is described as low in most study settings. The relative contribution of peripheral and central actions remains an active research question.
Semaglutide is a synthetic peptide of thirty-one amino acids that shares roughly ninety-four percent sequence identity with human glucagon-like peptide-1. Two substitutions resist enzymatic cleavage by dipeptidyl peptidase-4, and a fatty diacid side chain attached through a linker promotes binding to serum albumin. That albumin binding slows renal clearance and extends the circulating half-life from minutes to approximately one week. The structural changes are well established in the published literature. Whether the same modifications affect receptor signalling bias in ways that matter clinically remains an open question.
Pharmacological activity arises from agonism at the glucagon-like peptide-1 receptor, a G protein-coupled receptor expressed in the pancreas, the gastrointestinal tract, and the brainstem. Receptor activation raises intracellular cyclic adenosine monophosphate and enhances insulin release in a glucose-dependent manner, an effect that diminishes when blood glucose concentration is low. Other effects include slowed gastric emptying and hypothalamic satiety signalling. These pathways are described well. Receptor desensitisation rates across tissues, relative to the endogenous hormone, are still under investigation, and reported findings differ between laboratories.
The company that developed the compound filed it as a long-acting analogue, and it gained first approval in 2017 for type 2 diabetes. Later authorisations from several regulators extended the indication to chronic weight management, and the World Health Organization added the glucagon-like peptide-1 receptor agonist drug class to its model list of essential medicines in 2023. Production uses solid-phase peptide synthesis followed by side-chain conjugation and chromatographic purification. Supply constraints and cost differences across regions are well documented. Literature on long-term outcomes continues to grow, with many trials reporting surrogate endpoints rather than hard clinical endpoints.
== Epidemiology == Diabulimia is most common in women, and in people between 15 and 30 years old. Around 40% of men with type 1 diabetes may have skipped insulin injection at least once, and around 20% of women. Some studies have found that up to 60% of people with type 1 diabetes deliberately restrict insulin at some point. Many articles and studies further conclude that diabetic females have, on average, higher body mass index (BMI) than their nondiabetic counterparts. Girls and young adult women with higher BMIs are also shown to be more likely to have disordered eating behavior (DEB). Many authoritative articles show that preteen and teenage girls with type 1 diabetes have significantly higher rates of eating disorders of all types than do girls without diabetes.
In 1990, following cumulative $8.5 million donations by Bronx businessman Jack Resnick, Einstein's campus was designated as the Jack and Pearl Resnick Campus. Six years later, Einstein built a 10-story research complex, the Samuel H. and Rachel Golding Building. In 2002, the college opened the three-story Gruss Magnetic Resonance Research Center. Equipped with magnetic resonance equipment beyond conventional MRI, the center was one of six such facilities in the world upon opening. In 2008, Einstein opened a $225 million research complex, the Price Center. This expansion doubled the size of Einstein's campus to nearly 40 acres. Also that year, the college replaced its old logo—a "staid" portrait of Albert Einstein—with a stylized symbol that represents the helical structure of DNA, a notable spiral staircase on campus, and 'E' for Einstein. In 2024, Ruth Gottesman—a long-time professor at the medical school and head of the board of trustees—donated $1 billion to the school to make tuition free for all students in perpetuity. The contribution also stipulated that the college never change its name. The donation was one of the largest to any educational institution, and, according to The New York Times, likely the largest donation to any medical school.
== Nomenclature == Perfluorocarbons or PFCs, are organofluorine compounds with the formula CxFy, meaning they contain only carbon and fluorine. The terminology is not strictly followed and many fluorine-containing organic compounds are also called fluorocarbons. Compounds with the prefix perfluoro- are hydrocarbons, including those with heteroatoms, wherein all C-H bonds have been replaced by C-F bonds. Fluorocarbons includes perfluoroalkanes, fluoroalkenes, fluoroalkynes, and perfluoroaromatic compounds.
All mRNA templates used for mRNA display technology have puromycin at their 3' end. As translation proceeds, the ribosome moves along the mRNA template, and once it reaches the 3' end of the template, the fused puromycin will enter ribosome's A site and be incorporated into the nascent peptide. The mRNA-peptide fusion is then released from the ribosome (Figure 1). To synthesize an mRNA-peptide fusion, the fused puromycin is not the only modification to the mRNA template. Oligonucleotides and other spacers need to be incorporated along with the puromycin to provide flexibility and proper length for the puromycin to enter the A site. Ideally, the linker between the 3' end of an mRNA and the puromycin has to be flexible and long enough to allow the puromycin to enter the A site upon translation of the last codon. This enables the efficient production of high-quality, full-length mRNA-peptide fusions. Rihe Liu et al. optimized the 3'-puromycin oligonucleotide spacer. They reported that dA25 (a nucleotide sequence of 25 deoxyadenosine residues) in combination with a Spacer 9 (Glen Research), and dAdCdCP at the 5' terminus worked the best for the fusion reaction. They found that linkers longer than 40 nucleotides and shorter than 16 nucleotides showed greatly reduced efficiency of fusion formation. Also, when the sequence rUrUP was present adjacent to the puromycin, fusion did not form efficiently.

== Assessments == A December 2024 Gallup poll found that 54% of Americans thought Biden's presidency was below average or poor, 26% average, and 19% above average or outstanding. Gallup found that Americans largely offered negative assessments of Biden's presidency on economic, national, and international issues. Of 18 issue areas Gallup tracked, a majority of Americans said the U.S. lost ground in six, including the economy in general, immigration, and the country's position in the world. A plurality found that the U.S. declined in six other areas, including national infrastructure and energy, education, and trade relations with other countries. A plurality felt the U.S. made progress in only one indicator under Biden: the conditions of gay, lesbian, and transgender people. When Biden left office in 2025, journalists and many Democrats viewed his presidency as a failure due to age and health concerns, public frustration over inflation, and Harris's loss to Trump in the 2024 presidential election. The Guardian called the conclusion of Biden's presidency a "tragedy".
== Uses == In the eighteenth century English physician Thomas Fowler (1736–1801) utilized potassium arsenite to remedy a number of conditions including anemia, rheumatism, psoriasis, eczema, dermatitis, asthma, cholera, and syphilis. Furthermore, in 1865 the potential uses of potassium arsenite expanded as Fowler’s solution was used as the first chemotherapeutic agent to treat leukemia, however the effects were only temporary. Surprisingly enough, this specific use was inspired by potassium arsenite’s role in improving digestion and producing a smoother coat in horses. Potassium arsenite is also a key inorganic component of certain wood preservatives, rodenticides, insecticides, and herbicides.
By the end of the 15th century, spring-driven clocks appeared in Italy, and in Germany. Peter Henlein, a master locksmith of Nuremberg, was regularly manufacturing pocket watches by 1526. Thereafter, pocket watch manufacture spread throughout the rest of Europe as the 16th century progressed. Early watches only had an hour hand, the minute hand appearing in the late 17th century.

Filtering removes particulate matter that can block columns or scatter light. A 0.22 micrometre membrane is typical, and the filter material should be checked for peptide adsorption.
Peptides can bind to glass and plastic, so a fraction of the material leaves the solution. The effect is proportionally larger in dilute samples and can bias quantitative results.
Reverse-phase high-performance liquid chromatography is the most widely reported approach. Purity figures should always be quoted together with the wavelength, gradient, and integration parameters used.
It is a modified version of the natural hormone, with three amino acid changes and a fatty acid side chain added. These edits extend its half-life from minutes to about one week. The core receptor activity is retained.
