The Bradford assay uses the spectral properties of Coomassie brilliant blue G-250 to estimate the amount of protein in a solution. A protein sample is added to a solution of the dye in phosphoric acid and ethanol. Under the acid conditions the dye is normally a brownish colour but on binding to the protein the blue form of the dye is produced. The optical absorbance of the solution is measured at a wavelength of 595 nm. The dye is noted for its high level of sensitivity: 5 μg of protein can be detected. However, among the disadvantages of the method is its variability of color development with different proteins: the absorbance change per unit mass of proteins varies with the type of the protein. On binding to a protein, the negatively charged Coomassie brilliant blue G-250 dye molecule will give an overall negative charge to the protein. This property can be used to separate proteins or protein complexes using polyacrylamide gel electrophoresis under non-denaturing conditions in a technique called blue native PAGE. The mobility of the complex in the polyacrylamide gel will depend on both the size of the protein complex (i.e., the molecular weight) and the amount of dye bound to the protein. On 22 February 2026, El Mencho was killed by the Mexican military in Tapalpa, Jalisco. Widespread violence erupted in Mexico as a result of retaliation attacks from the CJNG. Mexico City-based security consultant David Saucedo informed CNN that due to El Mencho's son Rubén Oseguera González ("El Menchito") being jailed in the United States, the line of succession "has been broken," with no clear family successor. The arrest of El Menchito would leave only one brother and El Mencho's stepson Juan Carlos Valencia Gonzalez ("El Pelón"), though El Pelon lacked "influence among other cartel commanders." Saucedo stated that was "likely that one of four or five top commanders will emerge as the next leader," and named CJNG leaders Ricardo Ruiz Velasco ("El Doble R"), Audias Flores ("El Jardinero" or "The Gardener"), Hugo Mendoza Gaytan ("El Sapo") and a fourth figure – El Mencho's former head of security – about whom little is known, as the most likely candidates to succeed El Mencho. In addition it was soon afterwards revealed by El Pais that El Mencho's brother Abraham, who was arrested in Mexico in February 2025, was actually in fact still "in the hands of Mexican authorities." The U.S. Director of National Intelligence's (DNI) National Counterterrorism Center has stated that El Pelón was the "de facto second-in-command" to El Mencho, with $5 million bounties being issued each for El Pelon and El Jardinero.
== Molecular problem: fibrosis == When faced with pressure overload, the heart attempts to compensate with a number of structural alterations including hypertrophy of cardiomyocytes and increase of extracellular matrix (ECM) proteins. Rapid accumulation of ECM proteins causes excessive fibrosis resulting in decreased myocardial compliance and increased myocardial stiffness. The exact mechanisms involved in excessive fibrosis are not fully understood but there is evidence that supports involvement from local growth factors FGF-2, TGF-beta and platelet-derived growth factor. TGF-β1 plays an important role in cardiac remodelling through the stimulation of fibroblast proliferation, ECM deposition and myocyte hypertrophy. The increase in TGF-beta 1 expression in a pressure-overloaded heart correlates with the degree of fibrosis, suggesting TGF-beta 1 involvement in the progression from a compensated hypertrophy to failure. == Withdrawal == Withdrawal symptoms may occur during gradual or particularly abrupt withdrawal of tricyclic antidepressant drugs. Possible symptoms include: nausea, vomiting, abdominal pain, diarrhea, insomnia, headache, nervousness, anxiety, dizziness and worsening of psychiatric status. Differentiating between the return of the original psychiatric disorder and clomipramine withdrawal symptoms is important. Clomipramine withdrawal can be severe. Withdrawal symptoms can also occur in neonates when clomipramine is used during pregnancy. A major mechanism of withdrawal from tricyclic antidepressants is believed to be due to a rebound effect of excessive cholinergic activity due to neuroadaptations as a result of chronic inhibition of cholinergic receptors by tricyclic antidepressants. Restarting the antidepressant and slow tapering is the treatment of choice for tricyclic antidepressant withdrawal.
